Assessment of anti-inflammatory activity of Poria cocos in sodium lauryl sulphate-induced irritant contact dermatitis.

OBJECTIVE: In the present study, we evaluated the anti-inflammatory activity of Poria cocos (PoCo) on experimentally induced irritant contact dermatitis (ICD) in a repeated sodium lauryl sulphate (SLS) irritation model. METHODS: The anti-irritative effect of PoCo was evaluated with a visual score and quantified by non-invasive bioengineering methods, namely chromametry and trans epidermal water loss. Three concentrations of PoCo in base cream DAC (amphiphilic emollient; German pharmacopoeia) were tested in a 4-day repetitive irritation test using SLS. RESULTS: A statistically significant anti-inflammatory activity was observed for PoCo by all three methods when applied in parallel to the induction period of ICD. Application of PoCo after induction of ICD once a day for 5 days, starting just at the end of 4 days, was without any effect. CONCLUSION: An anti-inflammatory efficacy of PoCo on the elicitation phase of the ICD induced by repeated SLS test could be observed and quantified by three independent, non-invasive biophysical assessment parameters. This effect can be explained by its influence on pro-inflammatory enzymes, namely phospholipase A2.

Aloe Barbadensis

Effect of aloe gel on cell proliferation and synthesis of collage in cultured human skin fibroblast

Objective: To investigate the effect of aloe gel on cell proliferation and synthesis of collage in cultured human skin fibroblast. Methods: The primary human skin fibroblasts were isolated and cultured in vitro, which were treated with aloe gel in different concentration in exponential phase of cell growth. The hydroxyproline in the cultural medium was determined by alkaline hydrolysis. Cell cycle was measured by flow cytometry. The expression of pro-collagen type I was detected by immunohistochemistry. Results: Employing the flow cytometry technique we observed the cell cycle of S+G2/M stage and the proliferation index (PI) were increased (P<0.05), when the cells were stimulated by 5 %, 100% aloe gel. But there was no statistical difference in the cell cycle at the time point experiment (P>0.05). Compared with the control group the hydroxyproline in the cultural medium was increased (P<0.05), when the cells were stimulated by aloe gel. The expression of pro-collagen type T in vitro was increased (P<0.05) after the cells stimulated by 10%aloe gel. Conclusion: Aloe gel can enhance the growth of human skin fibroblast and the collage synthesis.

Shao Yao/Paeonia Lactiflora

Modern research showed that Bai Shao could increase the phagocytic percentage and index of celiac phagocyte in mice, promote cellular immunity, and restore the lowering amounts of antibody induced by cyclophosphane. It had anti-viral, anti-bacterial and anti-inflammatory effects. Total glucosides of Bai Shao could increase diastolic pressure and dp /dtmax, lower the heartbeat, and increase the blood pressure elevating effect of NA. In vitro tests showed that Bai Shao extracts could inhibit thrombosis. Glucosides of Shao Yao had spasmolytic and analgesic effect. It could also dosage-dependently lower the normal temperature of mice and rats. Total glucosides of Shao Yao could improve the sleep in different functional states in rats; lengthen the survival time under anoxic condition. Bai Shao extract could protect and liver, and paeoniflorin could significantly inhibit the peptic ulcer in rats induced by stress.

In clinic, Bai Shao is indicated in the treatment of viral hepatitis, acute bacillary dysentery, heart failure due to pulmonary heart disease, asthma, gastric and duodenal ulcer, bile reflux gastritis, chronic colonitis, constipation, senile chronic diarrhoea, diabetes, primary dysmenorrhea, infertility, whooping cough, migraine, trigeminal neuralgia and facial spasm.

Hong Hua /Carthamus Tinctorius

Modern research showed that low dosage of water extraction of Hong Hua could increase the blood flow volume in the coronary artery, slightly lower blood pressure and slightly increase the heart beat of the anaesthetized dog; while its ethanol extract could lower the blood pressure, lower the heart beat but have no influence over blood flow volume in the coronary artery. Hong Hua injection could dilate the peripheral vessels. In vitro tests showed that Hong Hua had anticoagulant effect and could inhibit the platelet aggregation. Hong Hua had exciting effect on isolated or in situ uterus in various animals. It had certain exciting effect on the respiratory centre in cerebral ischemic and anoxic rats. Hong had hypnotic and analgesic effects. Extractum of Hong Hua had inhibitory effect on Helicobacter pylori. Hong Hua could also prevent hepatocirrhosis induced by CCl 4.

In Clinic, Hong Hua can be used to treat refractory haemoptysis, lymphadenitis, phlebitis, furuncles, bedsore, angina pectoris in coronary heart disease, arrhythmia, ulcer and sudden deafness, etc.

Kinobeon A, purified from cultured safflower cells, is a novel and potent singlet oxygen quencher.

We recently reported that kinobeon A, produced from safflower cells, and suppressed the free radical-induced damage of cell and microsomal membranes. In the present study, we investigated whether kinobeon A quenches singlet oxygen, another important active oxygen species. Kinobeon A inhibited the singlet oxygen-induced oxidation of squalene. The second-order rate constant between singlet oxygen and kinobeon A was 1.15 x 10(10) M (-1) s (-1) in methanol containing 10% dimethyl sulfoxide at 37 degrees C. Those of alpha-tocopherol and beta-carotene, which are known potent singlet oxygen quenchers, were 4.45 x 10(8) M (-1) s (-1) and 1.26 x 10(10) M (-1) s (-1), respectively. When kinobeon A was incubated with a thermolytic singlet oxygen generator, its concentration decreased. However, this change was extremely small compared to the amount of singlet oxygen formed and the inhibitory effect of kinobeon A on squalene oxidation by singlet oxygen. In conclusion, kinobeon A was a strong singlet oxygen quencher. It reacted chemically with singlet oxygen, but it was physical quenching that was mainly responsible for the elimination of singlet oxygen by kinobeon A. Kinobeon A is expected to have a preventive effect on singlet oxygen-related diseases of the skin or eyes.

Inhibitory effects of active compounds isolated from safflower (Carthamus tinctorius L.) seeds for melanogenesis.

In order to develop a new skin-whitening agent, safflower (Carthamus tinctorius L.) seeds were evaluated for melanogenesis inhibitory activity and its active principles were identified following activity-guided isolation. The 80% aqueous methanol extract and ethyl acetate fraction from safflower seeds showed a significant inhibition for mushroom tyrosinase. Three active compounds, N-feruloylserotonin, N-(p-coumaroyl)serotonin, and acacetin, were isolated from the ethyl acetate fraction as the active principles. Compared with arbutin (IC50=0.223 mM), the IC50 values of these compounds were 0.023, 0.074, and 0.779 mM for N-feruloylserotonin, N-(p-coumaroyl)serotonin, and acacetin, respectively. It was also found that N-feruloylserotonin and N- (p-coumaroyl) serotonin strongly inhibited the melanin production of Streptomyces bikiniensis and B16 melanoma cells in comparison with a known melanogenesis inhibitor, arbutin (Roh et al, 2004).

Results of Evaluation:

30 subjects according with the criteria of acne (8 males and 22 females) took Acne Support for 30 days. The results showed that the number of acne reduced obviously, and the skin lesions were alleviated, 7 cases were markedly effective, 17 effective, the total effective rate was 80.00%. The results of routine blood and urine tests as well as biochemical indices were within normal range before and after the foretaste test. It could be believed that CSHC had obvious appearance caring and acne treating actions, and it had no obvious influence over subjects.

4.9 Abdominal ultrasonic B scan, EKG and fluoroscopy of chest: All were normal before and after the foretaste.

5. Conclusions

5.1 30 cases of acne subjects took Acne Support for 30 days according to the test requirements, and the number of acne reduced, skin lesions were alleviated. Among them, 7 cases were markedly effective, 17 effective, the total effective rate was 80.00%, which showed that Acne Support had obvious appearance caring and acne treating actions.

5.2 Acne Support could obviously improve the fatigue feeling, distending and pain in the hypochondriac region, dry throat and constipation of the acne subjects.

5.3 The changes between the results of serum testosterone and Corynebacterium acne before and after foretaste of Acne Support were not obvious.

5.4 Before and after foretaste of Acne Support, the testing results of HGB, RBC, WBC, serum total protein, albumin, GPT, GOT, carnine, carbamide, blood sugar, blood fat, and routine urine test were all within normal range, which showed that this product had no adverse effects on subjects.

5.5 No adverse reactions had been found during the foretaste of Acne Support.
